Forkhead box O3 (FOXO3) is a gene that encodes a protein that functions as a transcriptional activator. The protein transcriptionally initiates apoptosis in the absence of survival factors. Fusions, missense mutations, silent mutations, and frameshift deletions and insertions are observed in cancers such as intestinal cancer, skin cancer, and stomach cancer.

FOXO3 is altered in 1.29% of all cancers with breast invasive ductal carcinoma, lung adenocarcinoma, colon adenocarcinoma, pancreatic adenocarcinoma, and glioblastoma having the greatest prevalence of alterations [3].
