3'-phosphoadenosine 5'-phosphosulfate synthase 1 (PAPSS1) is a gene that encodes the sulfate donor cosubstrate for all sulfotransferase enzymes. Missense mutations, synonymous mutations, and nonsense mutations are observed in cancers such as colon cancer, endometrial cancer, and stomach cancer.

PAPSS1 is altered in 0.13% of all cancers with low-grade glioma, NOS, pancreatic adenocarcinoma, pilocytic astrocytoma, thyroid gland papillary carcinoma, and astrocytoma having the greatest prevalence of alterations [3].

The most common alterations in PAPSS1 are PAPSS1-BRAF Fusion (0.14%) and PAPSS1-TET2 Fusion (0.04%) [3].
