Overview

Generic Name(s):
lipid-encapsulated vincristine, liposomal vincristine, vincristine, liposomal, and vincristine liposome
NCI Definition [1]:
A liposomal formulation of Vincristine designed to reduce toxicity and improve efficacy. Vincristine binds irreversibly to microtubules and spindle proteins in S phase of the cell cycle and interferes with the formation of the mitotic spindle, thereby arresting tumor cells in metaphase. This agent also depolymerizes microtubules and may also interfere with amino acid, cyclic AMP, and glutathione metabolism; calmodulin-dependent Ca++ -transport ATPase activity; cellular respiration; and nucleic acid and lipid biosynthesis. (NCI)

Vincristine liposomal has been investigated in 10 clinical trials, of which 9 are open and 1 is closed. Of the trials investigating vincristine liposomal, 3 are phase 1 (2 open), 2 are phase 1/phase 2 (2 open), 2 are phase 2 (2 open), 2 are phase 3 (2 open), and 1 is no phase specified (1 open).

BCR-ABL1 Fusion, t(9;22)(q34;q11), and BCL2 Fusion are the most frequent biomarker inclusion criteria for vincristine liposomal clinical trials.

Acute lymphoblastic leukemia, anaplastic ependymoma, and B-cell acute lymphoblastic leukemia are the most common diseases being investigated in vincristine liposomal clinical trials [2].
