CTNNB1 (catenin (cadherin-associated protein), beta 1, 88kDa) is a gene that encodes catenin beta-1 protein (also known as ß-catenin). ß-catenin is part of a complex of proteins that form adherens junctions, which are important for the establishment and maintenance of epithelial cell layers by regulating cell growth and adhesion between adjacent cells (PMID: 17854762). Mutant ß-catenin has been implicated in the pathogenesis of several cancers including melanoma, colorectal cancer, hepatocelluar carcinoma, and ovarian cancer (PMID: 12781368).

CTNNB1 is altered in 3.20% of all cancers with uterine corpus neoplasm, colorectal adenocarcinoma, non-small cell lung carcinoma, melanoma, and hepatocellular carcinoma having the greatest prevalence of alterations [3].
