Low density lipoprotein receptor-related protein 6 (LRP6) is a gene that encodes a transmembrane low-density lipoprotein receptor protein that also functions as a co-receptor with Frizzled family proteins to transduce signals by Wnt proteins. Missense mutations, nonsense mutations, silent mutations, and frameshift deletions are observed in cancers such as endometrial cancer, intestinal cancer, and skin cancer.

LRP6 is altered in 0.01% of all cancers with astrocytoma and cutaneous melanoma having the greatest prevalence of alterations [3].

The most common alterations in LRP6 are LRP6 L1436F (3.57%), LRP6 Mutation (3.57%), and LRP6-ETV6 Fusion (0.00%) [3].
