N-myc downstream regulated 1 (NDRG1) is a gene that encodes a cytoplasmic protein that functions in stress responses, hormone responses, cell growth, and cell differentiation. The protein is also required for p53-mediated caspase activation and apoptosis. Fusions, missense mutations, nonsense mutations, silent mutations, and frameshift deletions and insertions are observed in cancers such as endometrial cancer, intestinal cancer, and skin cancer.

NDRG1 is altered in 0.03% of all cancers with prostate adenocarcinoma, angiosarcoma, clear cell renal cell carcinoma, dedifferentiated liposarcoma, and esophagogastric carcinoma having the greatest prevalence of alterations [3].
