Tumor necrosis factor, alpha-induced protein 3 (TNFAIP3) is a gene that encodes a protein that functions as a zinc finger protein and ubiquitin-editing enzyme. The protein inhibits NF-kappa B activation and TNF-mediated apoptotic death. Missense mutations, nonsense mutations, silent mutations, whole gene deletions, frameshift deletions and insertions, and in-frame deletions are observed in cancers such as hematopoietic and lymphoid cancers, intestinal cancer, and stomach cancer.

TNFAIP3 is altered in 1.65% of all cancers with colon adenocarcinoma, breast invasive ductal carcinoma, lung adenocarcinoma, diffuse large B-cell lymphoma, not otherwise specified, and endometrial endometrioid adenocarcinoma having the greatest prevalence of alterations [3].

The most common alterations in TNFAIP3 are TNFAIP3 Amplification (0.21%), TNFAIP3 Loss (0.16%), TNFAIP3 I207L (0.07%), TNFAIP3 T108A (0.07%), and TNFAIP3 E428Q (0.02%) [3].
